Glycyrrhizin micelle as a genistein nanocarrier: Synergistically promoting corneal epithelial wound healing through blockage of the HMGB1 signaling pathway in diabetic mice

The purpose of this study was to explore the feasibility targeting HMGB1 signaling pathway treat diabetic keratopathy with a dipotassium glycyrrhizinate-based micelle ophthalmic solution encapsulating genistein (DG-Gen), and evaluate whether these glycyrrhizinate (DG) micelles could synergistically enhance therapeutic effect encapsulated (Gen). An optimized DG-Gen fabricated Gen/DG weight ratio 1:15, formulation featured an encapsulation efficiency 98.96 ± 0.82%, average particle size 29.50 2.05 nm. observed have good in vivo ocular tolerance excellent corneal permeation, remarkably improve vitro antioxidant activity. Ocular topical application significantly prompted re-epithelialization nerve regeneration mice, efficacy might be due inhibition through down-regulation its receptors RAGE TLR4, as well inflammatory factor interleukin (IL)-6 IL-1β. In conclusion, data showed that is potential regulation target for treatment keratopathy, novel DG-micelle active agents such Gen cause blockage prompt wound healing.
